Anatomy and fructan distribution in vegetative organs of Dimerostemma vestitum (Asteraceae) from the campos rupestres Anais da ABC (AABC)

Among the compounds stored by plants, several functions are assigned to fructans, such as source of energy and protection against drought and extreme temperatures. In the present study we analyzed the anatomy and distribution of fructans in vegetative organs of Dimerostemma vestitum (Asteraceae), an endemic species from the Brazilian campos rupestres. D. vestitum has amphistomatic and pubescent leaves, with both glandular and non-glandular trichomes. In the basal aerial stem the medulla has two types of parenchyma, which differ from the apical portion. The xylopodium has mixed anatomical origin. Changes in pectins of the Xylopodium of Ocimum nudicaule from dormancy to sprouting Braz. J. Plant Physiol.

The thickened underground organ of Ocimum nudicaule is a tuber-like structure (xylopodium) that is dormant in winter and sprouts at the beginning of the spring. Changes in content of cell wall polysaccharides were shown to occur from dormancy to sprouting. Pectic polysaccharides of O. nudicaule were analyzed in relation to composition, molecular mass, and linkage structure in these two phenological phases. The pectin content was 33 % lower during sprouting when compared to dormancy. Changes were also observed in the molecular mass of the pectin fraction from dormancy to sprouting. Echinolaena inflexa (Poir.) Chase is a native C3 grass species with high biomass production and abundance in the cerrado comparable to Melinis minutiflora Beauv., a C4 forage grass of African origin, widespread in the cerrado, displacing some native herbaceous species. These species accumulate mainly starch, differently of Lolium multiflorum Lam. ssp. italicum Beck cv. Lema (ryegrass), a C3 temperate forage grass that accumulates fructans as the major storage carbohydrate in the vegetative tissues. A rota da embriogênese somática de Acca sellowiana, uma espécie arbórea frutífera com limitações quanto à propagação vegetativa, já foi estabelecida mas o número de embriões que restabelecem o crescimento após a fase cotiledonar é baixo. No presente trabalho, embriões somáticos em diferentes fases de desenvolvimento foram obtidos após tratamentos com 2,4-D e submetidos à condição de frio (4 °C) durante 20 dias, visando induzir a transformação de embriões somáticos em plântulas. Observou-se que 95% dos embriões zigóticos desenvolveram embriões somáticos quando tratados com 2,4-D de forma contínua (T1), enquanto que apenas 55% foram formados quando a auxina foi usada na forma pulsada (T2). Fructans and water suppression on intact and fragmented rhizophores of Vernonia herbacea BABT

In order to evaluate the involvement of fructans in drought tolerance, experiments were conducted using intact plants and excised rhizophores of Vernonia herbacea. The water content in rhizophores of intact plants was maintained for 30 days when plants were watered every 7 and 15 days, whereas plants remained alive up to 60 days without water. Total fructan, oligo- to polysaccharides ratio and mean molecular mass of polysaccharides increased in these plants indicating depolymerization of median size molecules. In apical fragments of rhizophores kept dry the oligosaccharides increased in relation to polysaccharides one day after excision in treated tissues. Produção de frutanos em calos e plântulas clonadas in vitro de Viguiera discolor Baker (Asteraceae) Acta Botanica

Viguiera discolor Baker é uma espécie nativa do cerrado, cuja sobrevivência está ameaçada pela invasão de espécies exóticas. Considerando sua elevada produção e a vasta aplicação de frutanos, o presente trabalho foi conduzido visando à obtenção in vitro dessa espécie e à prospecção desses carboidratos nessas condições. Sementes foram germinadas in vitro, em meio MS modificado, e após cinco semanas de incubação, nós caulinares foram isolados e incubados no mesmo meio adicionado de 0,5 mg L-1 de ANA, regenerando plantas uniformes, raízesnão espessadas, raízes tuberosas e estruturas semelhantes a calo (calo tipo1), formadas na região caulinar basal. Seeds of Caesalpinia echinata are tolerant to desiccation, keeping their viability up to 0.08 g H2O g-1 DM. When stored under laboratory conditions they lose their viability within one month, while under low temperatures germination ability is maintained during two years of storage. Here, we report on the variations of soluble carbohydrates, cyclitols and cyclitol galactosides in seeds of C. echinata during development and maturation, aiming for a better understanding of the involvement of these compounds in the maturation process and seed quality. Caesalpinia echinata Lam. (brazilwood) is an endangered tree species occurring along the eastern coast of Brazil from latitudes 5°56´S to 22°56´S. With such a range of distribution it is expected that temperature and daylength may affect plant development, which was examined in this study. One- and five-year-old saplings were grown under controlled photoperiods of 8 h, 12 h and 20 h, and in a greenhouse under natural light during 16 months. Woronin bodies in Penicillium janczewskii Zaleski BJM

Penicillium janczewskii Zaleski is an efficient microorganism for the production of extracellular inulinases and grows rapidly on medium containing sucrose or inulin as carbon source. Maintenance of this filamentous fungus on inulin medium induces secretion of large amounts of inulinases, but the resulting mycelium has thinner cell walls that easily collapse and break. Woronin bodies in hyphae of P. janczewskii grown on sucrose and inulin substrates were observed. No significant differences in the number, location, size and shape of Woronin bodies and level of plugging were observed in cultures of the fungus grown on the two carbon sources.
